A combined microsatellite multiplexing and boiling DNA extraction method for high-throughput parentage analyses in the Pacific oyster (Crassostrea gigas) ArchiMer

The analysis of parentage using microsatellite markers is of increasing importance, notably in aquaculture genetic research where communal rearing of mixed families can be used to reduce unwanted environmental variance. We present here an optimization of parental genotype assessment for larvae or adults of the Pacific oyster, Crassostrea gigas, using a multiplex system of three microsatellite loci. In conjunction with a simple DNA extraction protocol, this method enables high throughput analyses of parentage in C. gigas. Using this method, we successfully determined the parentage of 93% (1224 out of 1318) of the progeny in a factorial cross between 3 females and 10 males. A community of flower-visiting bees (Hymenoptera: Apoidea) in the coastal sand dunes of northeastern Brazil Biota Neotropica

We analyzed aspects of the community structure of Apoidea of a restricted area of dunes with restinga vegetation in Salvador, Bahia, Brazil. Standardized samples were taken for one year, 3 times a month from 6:00 a.m. to 6:00 p.m. A total of 3983 individuals (3770 females and 213 males) of 49 species, grouped into 13 morpho-functional types and belonging to five families, were collected from flowers in its majority. The fauna of Abaeté is composed by large solitary bees, best represented by Anthophoridae followed by Apidae; Halictidae; Megachilidae and Colletidae. A comparative study on fungal laccases immobilized on chitosan BABT

The phenoloxidase enzyme laccase from the cultures of the Pleurotus ostreatus and Botryosphaeria sp. and a commercial laccase from Aspergillus sp. were immobilized on chitosan of pharmaceutical degree by adsorption followed by crosslinking. Different immobilization conditions in relation to the granulometry of support and amount of enzymatic laccase extract used were tested, aiming at reaching high enzymatic activity with the immobilized enzyme. Two different substrates, ABTS and DMP, were used for the determination of enzymatic activity. To ignore the historical perspective of socio-economical phenomena while looking ahead to the future in an attempt to raise expectations and set new challenges, can prove a risky move when approaching the cooperative movement as a whole. This is particularly so in the case of the European agricultural cooperatives. We expect them to play a relevant role in the overall panorama of a globalized world. The aim of this article is to trace the origins of the agricultural cooperative movement in seven European countries (Belgium, Denmark, France, Italy, The Netherlands, Portugal and the United Kingdom) and discuss the coincidences and differences which account for common problems with a view to avoiding them in future joint ventures. A comparison of fitness between horses with different exercise history AgEcon

Anecdotal evidence suggests that a horse’s prior fitness level has an effect on the horse’s ability to return to that previous level of fitness. This trial aimed to test this assumption which would benefit horse trainers to create individual fitness programs for horses. Standardised exercise testing was used to compare the progress of two groups of horses during weeks three and seven of a 14-week training program. Group A had undergone a similar training program twelve months previously and Group B had not. Plasma lactate samples and heart rate monitoring were used to assess the levels of fitness of the horses. A comparison of growth and biochemical composition of Mytilus galloprovincialis (Lmk.) and Mytilus edulis (L.) on the West coast of Cotentin, Normandy, France 1999-2000 ArchiMer

The blue mussel, Mytilus edulis has been reared along the Normandy coast line since the 1960s. The gonadal cycle of this mussel species shows a sharp decrease in meat quality during the winter period after spawning. This decline in meat quality is so severe that sales have to be suspended from December to July. Another species of mussel, Mytilus galloprovincialis, which is reared in the lagoons along the French shore of the Mediterranean Sea has a different spawning cycle. An experiment was undertaken to study the meat quality of M. Galloprovincialis throughout the year after the spat were transferred from the Mediterranean Sea to the Normandy coast. A Comparison of Risk and Return Characteristics of Efficient Crop Portfolios for the Brown Soil Zones Saskatchewan and Mecklenburg, Germany AgEcon

Two efficient farms are constructed for the brown soils of Saskatchewan, Canada and for Mecklenburg, Germany based on producer panels. Both farms feature highly integrated cropping systems which take advantage of cropping synergies. However, farm risk is inherently different between the two because differences in 1) climate that gives rise to very different yield risk and cost structure, and 2) EU programs which offer fixed cash payments and stable sugar beet prices. As expected, risk is much higher for the Saskatchewan case farm - it has a chance of a negative cash flow of approximately one year in five. Organic farming is an alternative to conventional farming for providing sustainable crops with high export demand. This review analyses research findings on organic farming in Australia, with an emphasis on soil health. Several reports have indicated that organic farm management generally improves soil physical properties in Australia and elsewhere. Although low nitrogen (N) availability can constrain yields, organic farmers can improve N supply through legume green manures. Plant available phosphorus (P) is a more serious limiting factor in organic farming, particularly in Australia with naturally low P levels. A Comparison of X, Y, and Boomer Generation Wine Consumers in California AgEcon

This research shows that the wine market in the California is segmented by age. The wine-consumption behavior of the California wine consumer differs between the Generation X consumer and those in Generation Y and Baby Boomers. There are differences in demographics, purchasing attitudes, and purchasing behaviors among wine consumers from different generations. The findings from this research demonstrate that it is increasingly important to develop marketing strategies that are segmented for the target wine consumer. Generation Y are young wine consumers who are especially important for the industry because they offer an opportunity for growth–these younger generations are developing tastes now that are likely to last as they age.

A COMPARISON ON FARMERS’ PARTICIPATION IN FARMERS’ ORGANIZATIONS AND IMPLICATIONS FOR RURAL EXTENSION IN MINAS GERAIS AgEcon

This paper describes a comparison on farmers’ participation in farmers’ organisations and attempts to explain why they prefer to carry out production and marketing activities by themselves. It also examines the implications of the findings for rural extension and related services. A total of 122 interviews were conducted with different categories of farmers and with the extension workers. The comparative analysis of two case studies (Cruz Alta and Pantano) showed that socio-economic characteristics were not very significant in determining the level of farmers’ participation in farmers’ organizations in Minas Gerais. A complementary method for production of tetraploid Crassostrea gigas using crosses between diploids and tetraploids with cytochalasin B treatments ArchiMer

We present a new method to produce tetraploid Crassostrea gigas by cytochalasin B inhibition of polar body 2 expulsion in diploid females crossed with tetraploid males. This offers a means of direct introgression of genetic characters from selected diploid to tetraploid lines, avoiding a triploid step. Offspring larval ploidy shifted over time and depended on size, with tetraploids more frequent among the smaller larvae and triploids among the large. Viable tetraploids were found at 4 and 6 months, indicating the technique was successful. The possibility that gynogenesis occurred was tested by microsatellite analysis to confirm the presence of paternally inherited alleles. Following recent international oil price increases, there has been considerable interest in how this external factor can affect the South African economy. This paper reports results from a computable general equilibrium (CGE) analysis of an increase (up to 30 per cent) in international oil prices. Background information is provided, which puts the magnitude of the price variations in historical context. Coletas sistemáticas de abelhas em uma área restrita no Parque Estadual de Vila Velha, Paraná, no período de outubro de 2002 a outubro de 2003, resultaram em 1552 espécimes pertencentes a 181 espécies. Estas espécies estão distribuídas em 58 gêneros, 24 tribos e 5 subfamílias. As plantas visitadas correspondem a 113 espécies, em 72 gêneros e 38 famílias. Megachile com 20 espécies foi o gênero mais rico e Ceratina o gênero mais abundante dentre os gêneros nativos. Apis mellifera foi a espécie mais coletada, correspondendo a 28% do total de indivíduos, e Bombus atratus foi a espécie mais abundante dentre as abelhas nativas. cDNA microarray is an innovative technology that facilitates the analysis of the expression of thousands of genes simultaneously. The utilization of this methodology, which is rapidly evolving, requires a combination of expertise from the biological, mathematical and statistical sciences. In this review, we attempt to provide an overview of the principles of cDNA microarray technology, the practical concerns of the analytical processing of the data obtained, the correlation of this methodology with other data analysis methods such as immunohistochemistry in tissue microarrays, and the cDNA microarray application in distinct areas of the basic and clinical sciences.

The human adrenal cortex, involved in adaptive responses to stress, body homeostasis and secondary sexual characters, emerges from a tightly regulated development of a zone-specific secretion pattern during fetal life. Its development during fetal life is critical for the well being of pregnancy, the initiation of delivery, and even for an adequate adaptation to extra-uterine life. As early as from the sixth week of pregnancy, the fetal adrenal gland is characterized by a highly proliferative zone at the periphery, a concentric migration accompanied by cell differentiation (cortisol secretion) and apoptosis in the central androgen-secreting fetal zone. A Consistent Picture of Applied Protection Across the World AgEcon

In order to provide with a consistent assessment of protection across the world in 2001, the MAcMap-HS6 database includes ad valorem equivalent measure of tariff duties and tariff rate quotas for 163 countries and 208 partners, at the six-digit level of the Harmonized System (5,111 products), and accounting for all preferential agreements. We first describe the methodology used to compute and aggregate ad valorem-equivalent applied tariff duties, well suited for analytical purposes. Special emphasis is put in minimizing the endogeneity bias in the aggregation procedure, and in acknowledging structural differences in export specialization.
